چکیده

To the Editor: The presence of circulating tumor cells (CTC) in the peripheral blood of cancer patients has attracted a great deal of attention among investigators. The clinical significance of CTC has probably best been established in breast cancer, where it has been shown that the detection of CTC in metastatic patients correlates with early clinical relapse and patient death [1]. Gene expression profiling of CTC has been extended to other metastatic cancers including colorectal and prostate cancer [2,3]. Recently, a porous barrier density-gradient centrifugation system was developed that allows for enrichment of CTC from peripheral blood [4]. When coupled with the use of specific genes for real-time reverse transcriptase (RT)polymerase chain reaction (PCR), the buoyant density separation approach

Comparative Analysis of CD4+ and CD8+ T Cells in Tumor Tissues, Lymph Nodes and the Peripheral Blood from Patients with Breast Cancer

Background: CD4+ and CD8+ T cells are the main types of lymphocytes in cell-mediated immunity and play a central role in the induction of efficient immune responses against tumors. The frequencies of T cell subtypes in the peripheral blood and tumor tissues, and draining lymph nodes (dLN) can be considered as useful markers for evaluation of the immune system in cancers. Methods: In this study,...

Droplet Digital PCR Based Androgen Receptor Variant 7 (AR-V7) Detection from Prostate Cancer Patient Blood Biopsies

Androgen receptor splice variant V7 (AR-V7) was recently identified as a valuable predictive biomarker in metastatic castrate-resistant prostate cancer. Here, we report a new, sensitive and accurate screen for AR-V7 mRNA expression directly from circulating tumor cells (CTCs): We combined EpCAM-based immunomagnetic CTC isolation using the IsoFlux microfluidic platform with droplet digital polym...

Assessing the Diagnostic Value of Plasma-Free DNA in Prostate Cancer Screening

Background: Prostate cancer is the second form of cancer among men worldwide. For early cancer detection, we should identify tumors in initial stages before the physical signs become visible. The present study aims to evaluate the diagnostic value of cell-free DNA (cfDNA), its comparison with prostate-specific antigen (PSA) level in prostate cancer screening and also in patients with localized ...

Identification of Enhancer of Zeste Homolog 2 Expression in Peripheral Circulating Tumor Cells in Metastatic Prostate Cancer Patients: A Preliminary Study

PURPOSE Enhancer of zeste homolog 2 (EZH2), a kind of transcriptional repressor, is reportedly over-expressed in metastatic prostate cancer. In this study, we analyzed EZH2 mRNA in circulating tumor cells (CTCs) in peripheral blood as a biomarker in patients with metastatic prostate cancer. PATIENTS AND METHODS Ber-EP4 coated immunomagnetic beads were used to harvest CTCs, and mRNA was isolat...

Detection of androgen receptor mutations in circulating tumor cells in castration-resistant prostate cancer.

BACKGROUND Coding mutations in the AR (androgen receptor) gene have been identified in tissue samples from patients with advanced prostate cancer and represent a possible mechanism underlying the development of castration-resistant prostate cancer (CRPC). There is a paucity of tumor-derived tissue available for molecular studies of CRPC patients. Circulating tumor cells (CTCs) in the blood of C...
